Antibody Validation That Matches Real Lab Demands
At KinesisDx, we don’t stop at catalogue claims. Every antibody is validated in ways that matter for your experiment, not just in theory. Our validation framework aligns with industry best practices and global standards, including IWGAV (International Working Group for Antibody Validation) principles and real use case testing. This ensures the antibodies you choose deliver specific, reproducible, and publication ready results, whether you’re doing Western blots, IHC, flow cytometry, or other assays.
Our Validation Standards
We apply a rigorous, multi layered approach depending on target availability and the intended application.
Genetic Evidence (KO & KD Validation)
What it is
Knockout (KO): Target gene is deleted (e.g., via CRISPR).
Knockdown (KD): Target expression is reduced (e.g., via siRNA/shRNA).
Why it matters
A truly specific antibody should give a clear signal in wild type (WT) samples and no signal in KO or reduced signal in KD samples.
Where we apply it
Common in signaling and marker targets such as EGFR (cancer signaling), AKT1 (pathway biomarker), STAT2 (immune pathway), CD133 (stem cell marker), CD147/BSG (cell surface marker). This approach confirms that the antibody’s signal truly reflects the protein of interest, which is critical for publication, pathway analysis, and drug discovery.
Application Specific Validation
Antibodies are tested in the actual assays researchers use, not just one generic condition.
Western Blot (WB): Checks band size, specificity, and absence of non specific bands
Immunohistochemistry (IHC): Validates tissue staining patterns
Flow Cytometry: Ensures correct cell population detection
ELISA: Confirms ligand/antigen capture and signal dynamics
Immunofluorescence (IF): Visualizes sub cellular localization
ChIP/IP: Validates binding and protein complexes
For example, EGFR and AKT1 are validated in WB and IHC to confirm band specificity and tissue staining. STAT2 is validated in IF and flow to confirm immune activation patterns.
IWGAV Alignment
We follow the validation principles recommended by the International Working Group for Antibody Validation:
Genetic evidence
Orthogonal validation
Independent antibody validation
Recombinant protein validation
Tagged protein expression
Mass spectrometry cross validation
Not all methods are applicable for every antibody, but these guidelines give a structured, scientific backbone to our workflows.
Design Confidence Through AI Epitope Precision
Our antibodies are engineered using AI assisted epitope selection to focus reactivity on the most unique, biologically relevant protein regions. This enhances specificity and reduces off target binding compared with random or low stringency peptide antigens.
